目的:检测DNA甲基转移酶3a(DNA methltransferase 3a,DNMT3a)和组蛋白去乙酰化酶2(histone deacetylase 2,HDAC2)在人胃癌组织中的表达,分析其与临床特征的相关性。方法:应用Western blot方法检测30例配对人胃腺癌和癌旁组织中DNMT3a和HDAC2表达的差异;免疫组织化学方法检测90例胃腺癌患者手术切除癌组织中DNMT3a和HDAC2的表达水平,并分析其表达与临床病理学指标的关系。结果:Western blot检测结果显示癌组织中DNMT3a和HDAC2蛋白表达量明显高于癌旁组织(P<0.01);90例胃癌标本中,免疫组化结果显示DNMT3a阳性表达率为79%(71/90),HDAC2蛋白表达阳性率为86%(77/90),癌组织中DNMT3a表达强度与肿瘤分化程度和淋巴结转移相关(P<0.05),HDAC2表达强度与肿瘤浸润深度、病理学大体分型、TNM分期和淋巴结转移相关(P<0.05)。DNMT3a与HDAC2在胃癌组织中的表达呈正相关(P<0.05)。结论:DNMT3a与HDAC2是胃腺癌发生发展中的危险因素之一,一定程度上能反映胃腺癌的恶性程度,对预后的判断有一定价值,是一项有潜在价值的肿瘤相关标志物。
OBJECTIVE: To detect the expression of DNA methltransferase 3a (DNMT3a) and histone deacetylase 2 (HDAC2) in human gastric cancer and to analyze their correlation with clinical features. Methods: The expressions of DNMT3a and HDAC2 were detected by Western blot in 30 paired human gastric adenocarcinoma and paracancerous tissues. The expression of DNMT3a and HDAC2 in 90 cases of gastric adenocarcinoma was detected by immunohistochemistry Relationship between expression and clinicopathological parameters. Results: The results of Western blot showed that the expression of DNMT3a and HDAC2 in cancer tissues was significantly higher than that in adjacent non-cancerous tissues (P <0.01). The positive rate of DNMT3a expression in 79 gastric cancer specimens was 79% (71/90) ), The positive expression rate of HDAC2 protein was 86% (77/90). The expression intensity of DNMT3a in cancer tissues was correlated with tumor differentiation degree and lymph node metastasis (P <0.05). The expression intensity of HDAC2 and tumor invasion depth, TNM stage and lymph node metastasis (P <0.05). The expression of DNMT3a and HDAC2 in gastric cancer was positively correlated (P <0.05). Conclusion: DNMT3a and HDAC2 are one of the risk factors in the development of gastric adenocarcinoma. To some extent, DNMT3a and HDAC2 can reflect the malignant degree of gastric adenocarcinoma and have certain value in judging the prognosis. It is a potential tumor associated marker.
