目的探讨白细胞介素32(IL-32)在博来霉素(BLM)诱导的小鼠肺纤维化过程中各期的表达和意义。方法将72只雄性小鼠随机分为三组,每组24只。分别给予0.9%氯化钠注射液0.04ml(C组)和等量的BLM(5mg/kg)(B、A组),一次性气管内灌注。A组在BLM灌注后每天给予地塞米松5mg/kg腹腔注射。然后各组分别于第3、7、14和28天各处死6只小鼠,取肺组织,做病理切片、免疫组织化学染色,应用Westernblot法测定IL-32表达。结果B组肺组织病理切片按时间顺序呈现由肺泡炎至纤维化动态改变。B组肺组织中IL-32蛋白表达水平高于C组(P<0.01)和A组。结论IL-32在肺纤维化形成过程中高表达,可能在肺纤维化形成的早中期发挥重要作用。
Objective To investigate the expression and significance of interleukin 32 (IL-32) during bleomycin (BLM) -induced pulmonary fibrosis in mice. Methods 72 male mice were randomly divided into three groups of 24 rats. They were given 0.9% sodium chloride injection 0.04ml (group C) and equal amount of BLM (5mg / kg) (group B and group A) respectively. Group A was given dexamethasone 5mg / kg intraperitoneally daily after BLM infusion. Then 6 mice were sacrificed on the 3rd, 7th, 14th and 28th day in each group respectively. The lung tissues were taken for histopathology and immunohistochemical staining. The expression of IL-32 was detected by Western blot. Results The histopathological changes of lung tissue in group B were changed from alveolitis to fibrosis according to time sequence. The level of IL-32 protein in lung tissue of group B was higher than that in group C (P <0.01) and group A. Conclusion IL-32 is highly expressed in the process of pulmonary fibrosis and may play an important role in the early and middle stages of pulmonary fibrosis.
