目的建立成年人视网膜神经细胞的体外培养体系,研究细胞体外生长特性和结构特点。设计实验性研究。研究对象体外培养的成年人视网膜神经细胞。方法以成年人视网膜为研究对象,进行体外细胞培养;取培养7~10天的细胞行免疫组织化学鉴定;取培养10天的细胞行扫描电镜超微形态学检查。主要指标细胞形态,结构。结果在适宜的条件下,成年人视网膜神经细胞可以在体外培养并表现特有的结构特点和生长特性,免疫组化鉴定显示大部分细胞神经元特异性烯醇化酶(NSE)阳性,说明所培养细胞大部分为神经细胞。扫描电镜下观察贴壁伸展生长细胞主要为感光细胞、双极细胞、水平细胞等神经细胞和少量胶质细胞。结论在适宜的培养条件下,成年人视网膜神经细胞可以在体外进行培养并且表现出良好的生长活性。
Objective To establish an in vitro culture system of adult retinal nerve cells and study the growth characteristics and structural features of the cells in vitro. Design experimental research. The cells cultured for 7-10 days were identified by immunohistochemistry. The cells cultured for 10 days were examined by scanning electron microscopy. Results Under appropriate conditions, adult retinal neurons could be cultured in vitro and showed characteristic structural features and growth characteristics. Scanning electron microscopy observation of adherent growth cells mainly photoreceptor cells, bipolar cells, horizontal cells and other neural cells and a small amount of glial cells. Conclusion Under appropriate culture conditions, adult retinal neurons can be cultured in vitro and exhibit good growth activity.
