目的探讨来氟米特(LEF)对子宫内膜异位症模型大鼠内膜病灶的影响及对子宫内膜异位症可能的作用机制。方法运用自体移植法建立大鼠子宫内膜异位症模型成功的随机分LEF治疗组15只(A组:来氟米特35 mg·kg-1·d-1)、模型对照组14只(B组:生理盐水1 ml·kg-1·d-1)及未建模的15只大鼠为空白对照组(C组:生理盐水1 ml·kg-1·d-1)分别灌胃给药处理,运用流式细胞术检测血清中ICAM-1的水平变化,免疫组化法(SP)检测子宫内膜病灶中的RANTES表达情况。结果用药后LEF治疗组异位病灶体积明显小于模型对照组,ICAM-1、RANTES的表达低于模型对照组,差异均有统计学意义(P<0.05)。模型对照组ICAM-1、RANTES的表达高于空白对照组(P<0.01)。结论 LEF对子宫内膜异位症的进展有抑制行为。LEF治疗后大鼠ICAM-1、RANTES的表达明显降低,为来氟米特对子宫内膜异位症的治疗增加新的理论依据。
Objective To investigate the effect of leflunomide (LEF) on endometrial lesions in endometriosis model rats and the possible mechanism of endometriosis. Methods Fifteen rats were randomly divided into LEF group (A group: leflunomide 35 mg · kg -1 · d -1) and model control group 14 Group B: normal saline (1 ml · kg-1 · d-1) and untreated 15 rats were used as blank control group (Group C: saline 1 ml · kg-1 · d-1) The levels of ICAM-1 in serum were detected by flow cytometry. The expression of RANTES in endometrial lesions was detected by immunohistochemistry (SP). Results The volume of ectopic lesion in LEF treatment group was significantly lower than that in model control group, the expression of ICAM-1 and RANTES was lower than that of model control group, the difference was statistically significant (P <0.05). The expression of ICAM-1 and RANTES in the model control group was higher than that in the blank control group (P <0.01). Conclusions LEF can inhibit the progression of endometriosis. After LEF treatment, the expression of ICAM-1 and RANTES in rats decreased significantly, which added a new theoretical basis for the treatment of endometriosis with leflunomide.
