目的从基因水平分析河南地区汉族人群人类白细胞抗原HLA-DRB1等位基因型和频率,并了解其多态性分布状况。方法通过聚合酶链式反应-序列特异性引物(PCR-SSP)方法对1730名河南籍汉族健康人群进行DRB1等位基因分型。结果共检出13种DRB1等位基因型,频率最高的等位基因型为DRB1*15(18.12%),频率最低的等位基因型为DRB1*10(1.33%)。与湖北、广东和吉林三省比较,河南汉族人群DRB1各等位基因型频率分布与吉林省汉族人群最为相似,只有DRB1*03、DRB1*12、DRB1*13、DRB1*14和DRB1*16五个等位基因型频率存在显著统计学差异(P<0.05);与湖北省有8个等位基因型频率存在显著统计学差异(P<0.05);与广东省有9个等位基因型频率存在显著统计学差异(P<0.05)。结论在河南省与湖北、广东、吉林三省汉族人群DRB1位点多态性的比较中,河南汉族人群DRB1等位基因型频率分布与吉林省汉族人群最相似;高频率的DRB1*15等位基因型提示河南汉族人群可能易发生药物性肝损害。
Objective To analyze HLA-DRB1 alleles and frequencies of human leukocyte antigen (HLA-DRB1) in Henan Han population from the genetic level and to understand the distribution of HLA-DRB1 alleles. Methods DRB1 alleles were genotyped by polymerase chain reaction-sequence-specific primers (PCR-SSP) in 1730 Han healthy Han nationality. Results Thirteen DRB1 alleles were detected. The most frequent allele was DRB1 * 15 (18.12%) and the lowest allele was DRB1 * 10 (1.33%). Compared with the three provinces of Hubei, Guangdong and Jilin, the frequency distributions of DRB1 alleles in Han nationality in Henan province were the most similar to those in Han nationality in Jilin province. Only five DRB1 * 03, DRB1 * 12, DRB1 * 13, DRB1 * 14 and DRB1 * 16 There were significant differences in allele frequency (P <0.05) and 8 allele frequencies in Hubei Province (P <0.05). There were 9 allele frequencies in Guangdong Province Significant statistical difference (P <0.05). Conclusion The frequency distribution of DRB1 alleles in Henan Han population is the most similar to that of Han people in Jilin province in Han nationality in Henan province, Han nationality in Hubei province, Guangdong province and Jilin province. The high frequency DRB1 * 15 allele Hints Han Henan Han population may be prone to drug-induced liver damage.
