目的 :在昆虫细胞中表达乙型肝炎病毒 ( HBV)的表面抗原。方法 :利用已构建的含有 HBV S基因的重组杆状病毒 Bac- S,在昆虫细胞中进行表达 ,并用间接免疫荧光、ELISA和免疫印迹对表达产物进行检测。结果 :重组杆状病毒感染昆虫细胞后 ,可表达相应大小的 S蛋白 ,且该蛋白可被 HBs Ag特异性单抗 ( m Ab)所识别。结论 :利用杆状病毒表达系统 ,成功地表达了具有生物学活性的 HBV S蛋白。
Objective: To express hepatitis B virus (HBV) surface antigen in insect cells. Methods: The recombinant baculovirus Bac-S containing HBV S gene was constructed and expressed in insect cells. The expression products were detected by indirect immunofluorescence, ELISA and Western blotting. Results: The recombinant baculovirus could infect insect cells expressing the S protein of the corresponding size, and the protein could be recognized by HBs Ag specific McAb (m Ab). Conclusion: The biologically active HBV S protein was successfully expressed by baculovirus expression system.
