目的 探讨致康胶囊改善大鼠伤口愈合的作用与转化生长因子 β(TGF β)基因表达的关系。方法 取 2 0只雄性SD大鼠 ,在每个大鼠背部用外科方法剪去 2块 1.8cm× 1.8cm的全层皮肤 ,造成两个全皮层开放伤口。将每一只大鼠的两个伤口随机分为正常对照组伤口 (伤口总例数 =2 0个 )、致康胶囊治疗组伤口 (伤口总例数 =2 0个 )。术后连续 14d在致康胶囊治疗组伤口局部应用两粒去掉胶囊壳的致康胶囊粉剂 (80 0mg/伤口 )换药 ,在正常对照组伤口只进行常规换药。伤后 5、7、10、14d测定伤口愈合指数 ,伤后 5、7、10d检测伤口组织中TGF βmRNA表达水平。 结果 伤后 5、7、10、14d致康胶囊治疗组伤口愈合指数均高于正常对照组伤口 ,差异有显著性意义(P <0 .0 5 ) ;伤后 5、7d致康胶囊治疗组伤口组织中TGF βmRNA水平量均高于正常对照组伤口 ,差异有显著性 (P <0 .0 5 ) ;伤后 10d两组间TGF βmRNA水平量差异无显著性(P >0 .0 5 )。结论 致康胶囊促大鼠伤口愈合与其增强伤口组织TGF β基因表达相关
Objective To investigate the effect of Zhikang capsule on wound healing in rats and its relationship with the expression of transforming growth factor β (TGFβ) gene. Methods Twenty male Sprague-Dawley rats were used. Two full-thickness 1.8cm × 1.8cm skin were cut surgically in the back of each rat, resulting in two full-cortex open wounds. The two wounds of each rat were randomly divided into normal control group (the total number of wounds = 20), and the wounds of the treated group (total number of wounds = 20). Two consecutive capsules of Zhikang capsules (80 mg / wound) were applied to the wounds of Zhikang capsule group after operation for 14 consecutive days, and only the conventional dressing was applied to the wounds in the normal control group. The wound healing index was measured 5, 7, 10 and 14 days after injury. The expression of TGFβ mRNA in wound tissues was detected 5, 7 and 10 days after injury. Results The index of wound healing of Zhikang capsule treated group was significantly higher than that of normal control group on the 5th, 7th, 10th, 14th day after injury (P <0.05) The level of TGF-βmRNA in the wound tissue was higher than that in the normal control group (P <0.05). There was no significant difference in the level of TGF-βmRNA between the two groups at 10d after injury (P> 0.05) . Conclusion Zhikang capsule can promote the wound healing in rats and increase the expression of TGFβ gene in wound tissues
