目的观察Beagle犬牙槽隔减阻牙周膜牵张(periodontal ligament distraction,PDLD)快速移动牙齿压力侧牙周组织中RANKL和OPG的表达变化。方法 8只纯种Beagle犬随机分为四组,每只犬下颌左右第一前磨牙随机分为PDLD侧和常规方法正畸牙齿移动对照侧。测量相同加力时间内移动牙的移动距离;取标本,HE染色观察移动牙压力侧牙周组织的形态学变化;TRAP染色观察压力侧牙周组织中破骨细胞数目的变化;免疫组化染色观察移动牙压力侧牙周组织中RANKL、OPG的变化。结果加力后,PDLD侧移动牙压力侧牙周组织中TRAP阳性破骨细胞数明显增加,在加力2周时达峰值。PDLD侧移动牙压力侧牙周组织中RANKL和OPG含量均有升高,RANKL/OPG含量比值在加力2周时最高。结论与常规正畸牙齿移动相比,在牙槽隔减阻牙周膜牵张移动牙齿过程中RANKL、OPG的表达和RANKL/OPG含量比值变化更显著,与移动牙压力侧牙周组织的快速改建有关。
Objective To observe the expression changes of RANKL and OPG in the periodontal tissues of the beagle dogs with periodontal ligament distraction (PDLD) rapid pressure tooth movement. Methods Eight purebred Beagle dogs were randomly divided into four groups. The first premolar around the mandible of each dog was randomly divided into PDLD side and orthodontic tooth movement control side. The movement distance of the moving tooth during the same force application time was measured. The morphology of the periodontal tissue in the pressure side of the moving tooth was observed by HE staining. The number of osteoclasts in the periodontal tissue on the pressure side was observed by TRAP staining. The changes of RANKL and OPG in the periodontal tissues on the pressure side of the moving tooth were observed. Results After the application of force, the number of TRAP positive osteoclasts in the periodontal tissues on PDLD side significantly increased and reached its peak at 2 weeks after force application. The contents of RANKL and OPG in the periodontal tissues on the moving side of PDLD increased, and the ratio of RANKL / OPG was the highest at 2 weeks. Conclusion Compared with conventional orthodontic tooth movement, the ratio of RANKL, OPG and RANKL / OPG in alveolar fastigial periodontal membrane is more significant than that in orthodontic tooth movement.
