Prostate cancer was initially androgen-dependent, with tumors shrinking upon drug or surgical testicular castration but eventually turning to androgen-independent growth. The existing explanation is that the amplification or mutation of the androgen receptor (AR) hormone binding domain changes the binding function of AR, such as the ability of AR to bind to estradiol. AR can also be activated by insulin-like growth factor-1 (IGF-1), epidermal growth factor (EGF) and keratinocyte growth factor (KGF) in a ligand-independent manner.
