目的观察附子生物碱与甘草活性物质(三萜皂苷和黄酮等)组合对大鼠佐剂性关节炎(AA)模型的抗炎效应,研究两者配伍的抗炎作用机制,探讨两者配伍的意义。方法设对照组、模型组、附子组(即附子生物碱组)、附子甘草配伍组(即附子生物碱与甘草活性物质配伍组)、雷公藤组,观察各组大鼠足肿胀程度和关节炎指数(AI)变化;检测大鼠血清和关节浸液内细胞因子如白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)及前列腺素E2(PGE2)水平;用RT-PCR检测踝关节组织IL-1β基因表达;观察踝关节组织病理改变。结果与对照组比较,AA模型组大鼠关节肿胀严重,AI明显升高,血清和关节浸液内IL-β、TNF-α、PGE2水平显著升高,踝关节组织IL-1基因表达明显增加,并发生明显病理改变;与模型组相比,附子组、附子甘草配伍组大鼠关节肿胀得到改善,AI降低,血清和关节浸液内IL-1β、TNF-α、PGE2水平降低,踝关节组织IL-1β基因表达减弱,病理组织改善,且附子甘草配伍组优于附子组。结论附子生物碱与甘草活性物质配伍组合能有效抗炎,缓解AA症状,改善AA大鼠免疫功能和组织结构,两者配伍作用优于单用附子生物碱,体现了附子生物碱、甘草活性物质是附子甘草两者配伍抗炎效应的重要物质基础。
Objective To observe the anti-inflammatory effect of the combination of aconite alkaloids and licorice root active substances (triterpene saponin and flavonoids) on the rat model of adjuvant arthritis (AA). The levels of IL-β, TNF-α and PGE2 in serum and joint infiltration were significantly increased, and the expression of IL-1 gene in ankle joint was significantly increased , And significant pathological changes occurred. Compared with the model group, the swelling of the joint in the aconite group and the aconite-licorice compatibility group was improved, the AI was decreased, the levels of IL-1β, TNF-α and PGE2 in the serum and joint infiltration decreased, Tissue IL-1β gene expression decreased, pathological organization improved, and aconite licorice compatibility group than aconite group. Conclusions The combination of aconite alkaloids with licorice root active ingredients can effectively reduce the symptoms of AA and improve the immune function and structure of AA rats. The combination of alkaloids and licorice root active ingredients is better than that of aconite alkaloids alone.
