目的研究四氯二苯并二叶恶英(TCDD)诱导HepG2肝癌细胞、肺癌A549、SPC鄄A1细胞的细胞凋亡。方法分别用0.01、0.10、1.00μmol/LTCDD对3种细胞染毒24h,再细胞爬片和HE染色,光学显微镜下观察细胞形态。结果细胞凋亡可在0.01、0.10、1.00μmol/LTCDD处理后24h出现。表现为细胞固有形态丧失,核染色质固缩、碎裂,出现凋亡小体,并随染毒浓度加大而更明显。SPC鄄A1细胞的凋亡率分别为9.56%,16.8%,21.7%;A549的凋亡率分别为7.7%,13.6%,19.4%;HepG2的凋亡率分别为8.5%,14.5%,30%,均高于对照组,3组癌细胞凋亡率与对照组相比,差异有统计学意义(P<0.05)。结论TCDD可能诱导此3种细胞发生凋亡。
Objective To study the apoptosis of HepG2 hepatocarcinoma cells, lung cancer A549 and SPC-A1 cells induced by tetrachlorodibenzo-p-dioxins (TCDD). Methods The three kinds of cells were exposed to 0.01,0.10,1.00μmol / L DCD for 24 hours respectively. Cell slides and HE staining were used to observe the cell morphology under light microscope. Results Apoptosis occurred at 24h after treatment with 0.01,0.10,1.00μmol / LTCDD. Manifested as the loss of cell intrinsic form, nuclear chromatin condensation, fragmentation, apoptotic bodies appear, and with the increase in the concentration of the more obvious. The apoptotic rates of SPC-A1 cells were 9.56%, 16.8% and 21.7%, respectively. The apoptosis rates of A549 cells were 7.7%, 13.6% and 19.4%, respectively. The apoptosis rates of HepG2 cells were 8.5%, 14.5% and 30% , Which were higher than that of the control group. The apoptosis rate of cancer cells in 3 groups was significantly different from that in control group (P <0.05). Conclusion TCDD may induce apoptosis of these three kinds of cells.
