鼠类实验性血吸虫病有骨髓、血液和组织内嗜酸性粒细胞(EOS)增多的特征。病程超过90天的慢性血吸虫病,宿主血液中EOS稍升高,而组织中EOS的增多十分显著。已建立一实验模型,即在患慢性血吸虫病小鼠腹腔内植入小玻片以引起炎症,该鼠同时有外周EOS的增殖,并证明此增殖依赖于游走的巨噬细胞(Mφ)。EOS的增生一般认为白细胞介素5(IL-5)所控制。本研究是为了确定血吸虫病时EOS的增殖型式以及在骨髓中EOS生成与外周EOS增多的关系,阐明游走到玻片植入物上的细胞是否分泌EOS增殖因子。取1月龄C3H/HeJ小鼠感染曼氏血吸虫尾蚴30条,于45~50天(急性期)及90~120天(慢性期)分别进行研究,设正常对照组。用McCoy 5A培养基冲洗股骨腔
Murine experimental schistosomiasis is characterized by an increase in bone marrow, blood and tissue eosinophils (EOS). Chronic schistosomiasis more than 90 days duration, EOS slightly increased in the host blood, and tissue EOS increased significantly. An experimental model has been established in which small slides are implanted intraperitoneally in mice with chronic schistosomiasis to induce inflammation. The mice also have peripheral EOS proliferation and demonstrate that this proliferation is dependent on migratory macrophages (Mφ). EOS hyperplasia is generally considered interleukin 5 (IL-5) control. The purpose of this study was to determine the proliferative patterns of EOS during schistosomiasis and the relationship between EOS production and peripheral EOS in the bone marrow to elucidate whether cells migrating to the slide implants secrete EOS growth factors. 30-month-old C3H / HeJ mice were infected with 30 S. cercariae Schistosomes, and were studied in 45-50 days (acute phase) and 90-120 days (chronic phase) respectively.
