目的 用纤维蛋白原转化成的纤维蛋白刺激培养的血管平滑肌细胞,通过细胞计数和3H-胸腺嘧啶参入的方法,测定平滑肌细胞的增殖情况,并用 Northern 杂交的方法观测其c-fos、c-m ycm RNA水平。结果 纤维蛋白可明显促进平滑肌细胞的增殖,其作用呈浓度依赖关系。静止状态细胞,c-fos、c-m yc 的m RNA 呈低表达,纤维蛋白刺激后,二者表达均明显增加。结论 纤维蛋白刺激平滑肌细胞的增殖与促进c-fos、c-m yc原癌基因的表达有关
OBJECTIVE: To investigate the proliferation of vascular smooth muscle cells (VSMCs) cultured in vitro by using fibrin converted from fibrinogen. The proliferation of VSMCs was measured by cell counting and 3H-thymidine incorporation. The c-fos and c-m ycm RNA level. Results Fibrin could significantly promote the proliferation of smooth muscle cells in a concentration-dependent manner. The m RNA of c-fos and c-m yc was low expression in quiescent cells, and the expression of m RNA was significantly increased after fibrin stimulation. Conclusion Fibrin stimulates the proliferation of smooth muscle cells and promotes the expression of c-fos and c-mcc oncogenes
