作者在小鼠和家兔中对作为人菌苗抗原破伤风类毒素(TT)和白喉类毒素(DT)佐剂的、由二氧乙烯鲸蜡醚、胆固醇和油酸组成的非磷脂脂质体进行了评估.分别将TT和DT混合或包被在脂质体、脂质体-单磷酸脂质A(MPL)/鲨烯和脂质体-鲨烯中制成不同的脂质体抗原制剂,以磷酸铝吸附的抗原、结合弗氏佐剂的抗原及可溶性抗原为对照.分别用上述抗原免疫小鼠(2剂TT)和家兔(3剂DT).于首剂TT免疫后4、6、10和15周及每剂DT免疫后2周采血.用ELISA法测定抗-TT IgG、IgG亚类和IgM抗体及抗-DT IgG抗体.用毒素中和试验测定小鼠血清中破伤风抗毒素水平,用Vero细胞测定兔血清白喉抗毒素.
The authors tested non-phospholipid lipids composed of ethylene oxide cetyl ether, cholesterol, and oleic acid in mice and rabbits as adjuvants to human vaccine antigens tetanus toxoid (TT) and diphtheria toxoid (DT) The TT and DT were mixed or coated separately in liposomes, liposome-monophosphate lipid A (MPL) / squalene and liposomal-squalene to make different liposome antigens The preparations were immunized with aluminum phosphate adsorbed antigen, antigen combined with Freund’s adjuvant and soluble antigen as controls, respectively.The mice were immunized with the above antigen (2 doses of TT) and rabbits (3 doses of DT) , 6, 10, and 15 weeks and each dose of DT after 2 weeks of immunization. Anti-TT IgG, IgG subclasses and IgM antibodies and anti-DT IgG antibodies were determined by ELISA. Haemotoxin levels were measured in rabbit serum diphtheria antitoxin using Vero cells.
