目的:探讨丹皮酚(Pae)单药及联合顺铂(CDDP)对人卵巢癌细胞A2780s的增殖抑制,凋亡诱导作用及对细胞周期分布的影响。方法:以不同浓度的Pae、CDDP和两药联合作用于人卵巢癌细胞A2780s,采用MTT法测定药物对体外培养的人卵巢癌细胞A2780s增殖的抑制作用,应用两药相互作用指数(CDI)进行联合用药分析,流式细胞仪测定细胞凋亡情况及细胞周期分布的变化。结果:Pae(7.81-250mg/L)和CDDP(0.625-20mg/L)单独应用均对人卵巢癌细胞A2780s有抑制作用,呈现剂量-时间依赖效应关系。Pae在62.5mg/L与CDDP在5mg/L联合应用时协同作用最为显著(CDI=0.615)。流式细胞仪检测联合用药组(Pae62.5mg/L+CDDP5mg/L)凋亡率(44.9%)明显高于对照组和单独用药组。结论:Pae与CDDP均能抑制人卵巢癌细胞A2780s的增殖并诱导其发生凋亡,联合应用具有明显协同抗肿瘤作用。
Objective: To investigate the effects of paeonol and cisplatin (CDDP) on the proliferation, apoptosis and cell cycle distribution of human ovarian cancer cell line A2780s. Methods: PaO2, CDDP and two drugs were used in combination on human ovarian cancer cell line A2780s. MTT assay was used to determine the inhibitory effect of the drug on the proliferation of human ovarian cancer cell line A2780s cultured in vitro. The drug interaction index (CDI) Analysis of combination therapy, flow cytometry determination of apoptosis and cell cycle distribution changes. Results: Pae (7.81-250mg / L) and CDDP (0.625-20mg / L) alone could inhibit A2780s in a dose-dependent manner. Pae at 62.5mg / L and CDDP at 5mg / L synergistic combination of the most significant (CDI = 0.615). The apoptotic rate (44.9%) of the combined drug group (Pae62.5mg / L + CDDP5mg / L) by flow cytometry was significantly higher than that of the control group and the drug alone group. Conclusion: Both Pae and CDDP can inhibit the proliferation of human ovarian cancer cell line A2780s and induce their apoptosis. The combination of Pae and CDDP has obvious synergistic antitumor effect.
