目的:探讨虎栀合剂(Hu Zhi Mixture,HZM)防治α-异硫氰酸萘酯(α-maphthy1 Isothiocyanate,ANIT)致大鼠急性肝损伤的效应及机制。方法:将大鼠随机分为4组:正常对照组、造模对照组、虎栀合剂治疗组、泼尼松对照组。检测大鼠在造模48h后,血清中肝功能、肿瘤坏死因子-α(TNF-α)、肝组织中的丙二醛(MDA)、超氧化物歧化酶(SOD)的含量,光镜下观察肝脏组织学改变。结果:HZM能明显改善造模引起的肝组织学及肝功能变化,降低TNF-α、MDA水平,而SOD含量显著增高。泼尼松对照组能部分改变各项指标。结论:HZM防治ANIT致大鼠急性肝损伤切实有效。其作用机制可能与其拮抗作用,减少TNF-α、MDA的产生,提高SOD活性有关。
Objective: To investigate the effect and mechanism of Hu Zhi Mixture (HZM) on acute liver injury induced by α-maphthy1 Isothiocyanate (ANIT) in rats. METHODS: Rats were randomly divided into 4 groups: normal control group, model control group, Huqi Mixture treatment group, and prednisone control group. The serum levels of hepatic function, tumor necrosis factor-α (TNF-α), malondialdehyde (MDA), and superoxide dismutase (SOD) in liver tissue were measured after 48 hours of modeling. Observe liver histological changes. RESULTS: HZM could significantly improve the hepatic histology and liver function changes caused by modeling, reduce the levels of TNF-α and MDA, and increase the content of SOD. The prednisone control group can partially change the indicators. Conclusion: HZM is effective and effective in preventing acute liver injury induced by ANIT in rats. Its mechanism of action may be related to its antagonistic effect, reducing the production of TNF-α, MDA, and increasing SOD activity.
