目的:通过建立基因表达谱,研究透明细胞癌和浆液性囊腺癌之间差异表达基因。方法:利用cDNA芯片建立和分析两者基因表达谱,并对透明细胞癌中上调表达的膜联蛋白Ⅳ,在组织和细胞中进行了定量PCR以及免疫组织化学和Western blot验证。结果:卵巢透明细胞癌和卵巢浆液性囊腺癌基因表达谱比较,差异2倍以上基因共有82个,25个表达上调,57个表达下调。其中有功能的上调基因7个,下调基因12个。组织和细胞中膜联蛋白Ⅳ在mRNA及蛋白水平上的表达与基因芯片结果一致。结论:通过基因芯片可以建立卵巢透明细胞癌和浆液性囊腺癌的差异表达基因谱,膜联蛋白Ⅳ在卵巢透明细胞癌中上调表达可能具有重要意义。
OBJECTIVE: To study differentially expressed genes between clear cell carcinoma and serous cystadenocarcinoma by establishing gene expression profiles. Methods: Gene expression profile was established and analyzed by cDNA microarray. Annexin Ⅳ was up-regulated in clear cell carcinoma, and quantitative PCR and immunohistochemistry and Western blot were performed in tissues and cells. Results: The gene expression profiles of ovarian clear cell carcinoma and ovarian serous cystadenocarcinoma were compared. There were 82 genes with more than 2-fold difference, 25 were up-regulated and 57 were down-regulated. There are 7 functional up-regulated genes and 12 down-regulated genes. The expression of Annexin IV at mRNA and protein level in tissues and cells was consistent with the result of gene chip. Conclusion: Gene expression profiles of clear cell carcinoma and serous cystadenocarcinoma can be established by gene chip. Upregulation of annexin Ⅳ in clear cell carcinoma of the ovary may be of great significance.
