目的:探讨再生障碍性贫血(aplastic anermia,AA)大鼠模型的骨髓中凋亡因子CD34~+、Fas(CD95)、TNF-α表达与中医证型的关系。方法:将60只Wistar雄性大鼠随机分为正常组、模型组、心脾两虚组、脾肾阳虚组、肝肾阴虚组,每组12只。采用马利兰丙酮提取液腹腔注射制备大鼠AA模型,然后按相关文献方法加重模型大鼠的心脾两虚、脾肾阳虚、肝肾阴虚症状。检测各组大鼠的白细胞(WBC)、红细胞(RBC)、血小板(PLT)、血红蛋白(Hb)水平及其骨髓CD34~+、Fas、肿瘤坏死因子-α(TNF-α)表达情况。结果:1与正常组比较,其余各组大鼠的Hb、RBC、WBC、PLT水平均显著降低(P<0.05),提示造模成功;与模型组比较,肝肾阴虚组大鼠的Hb、RBC、WBC、PLT水平降低更加显著(P<0.05)。2与正常组比较,其余各组大鼠的骨髓CD34~+表达均明显下调(P<0.05),Fas、TNF-α表达明显上调(P<0.05);与模型组比较,肝肾阴虚组大鼠的骨髓CD34~+下调更加显著(P<0.05),Fas、TNF-α表达上调更加显著(P<0.05)。结论:肝肾阴虚型再生障碍性贫血大鼠的病情更加严重,这与临床中再生障碍性贫血“阳虚易治,阴虚难调”的理论基本一致,同时也为再障的中医辨证分型提供了一定的客观化依据。
Objective: To investigate the relationship between the expression of CD34 +, Fas (CD95) and TNF-α in the bone marrow of the rat model of aplastic anermia (AA) and the TCM syndromes. Methods: Sixty Wistar male rats were randomly divided into normal group, model group, xinpi deficiency group, spleen and kidney yang deficiency group, liver and kidney yin deficiency group, 12 rats in each group. The rat model of AA was prepared by intraperitoneal injection of acetone extract of Malilan, and then the symptoms of deficiency of spleen and kidney, yang deficiency of the spleen and kidney and liver-kidney yin deficiency were exacerbated according to the related literatures. The WBC, RBC, PLT, Hb and the expression of CD34 +, Fas and TNF-α in the bone marrow of each group were detected. Results: 1 Compared with the normal group, the levels of Hb, RBC, WBC and PLT in the other groups were significantly decreased (P <0.05), suggesting that the modeling was successful. Compared with the model group, Hb , RBC, WBC, PLT levels decreased more significantly (P <0.05). Compared with normal group, the expression of CD34 + in bone marrow of the other groups were significantly decreased (P <0.05), and the expressions of Fas and TNF-α were significantly increased (P <0.05). Compared with model group, The bone marrow CD34 ~ + was down-regulated more significantly in rats (P <0.05), and the expression of Fas and TNF-α was up-regulated more significantly (P <0.05). Conclusion: The condition of liver and kidney yin deficiency aplastic anemia is more serious, which is consistent with the theory of aplastic anemia in clinical practice, TCM syndromes provide a certain objective basis.
