In semi-solid agar culture, human peripheral blood mononuclear cells can be stimulated to form lymphocyte colonies. This culture technique is believed to be useful in studying the regulation of mitogen-induced lymphocyte responses in proliferative systems. These agar cultures are typically carried out in plates at a volume of 1 ml minimum. Recently, the application of glass capillaries to culture human TL-CFU and murine B lymphocyte colonies in 30 μl of agar medium has been described. One of the advantages of glass capillary culture is that it saves material, and since the colonies are linearly arranged at the bottom of the capillary, colony counting takes less time. The authors describe a glass capillary human TL-CFU agar culture system that is fast and repeatable without the addition of serum.
