目的:考察开窍药对缺血缺氧PC12细胞的保护作用及其部分机制。方法:采用连二亚硫酸钠(Na2S2O4)造成PC12细胞拟缺血缺氧损伤模型,用MTT比色法,考察对PC12细胞活力的影响,并按试剂盒要求,采用分光光度法,测定细胞内Ca2+含量与培养上清中NO含量。结果:与模型组比较,麝香、冰片、苏合香、安息香以及麝香与冰片各配比组均能显著升高细胞MTT值(P<0.05);冰片各剂量组、麝香与冰片1:1、1:2、1:3组,安息香37.5μg/100μl组,苏合香12.5μg/100μl组的细胞Ca2+含量均明显低于模型组(P<0.05);麝香、冰片、苏合香、安息香以及麝香与冰片各配比组的细胞NO含量均显著高于模型组(P<0.05)。结论:开窍药以及麝香与冰片各配比组能有效保护缺糖缺氧损伤的PC12细胞,其机制可能与提高细胞上清NO含量,进而降低细胞Ca2+的过度内流,减轻细胞内钙超载相关。
Objective: To investigate the protective effect and its mechanism of Caoki Medicine on hypoxic-ischemic rat PC12 cells. Methods: The hypoxic-ischemic injury model of PC12 cells was induced by sodium dithionite (Na2S2O4). The effect of Na2S2O4 on the viability of PC12 cells was investigated by MTT colorimetric assay. The content of Ca2 + in cells was determined by spectrophotometry And culture supernatant NO content. Results: Compared with the model group, the MTT values of all the groups of musk, borneol, strychnine, benzoin and musk and borneol increased significantly (P <0.05) (P <0.05). The contents of Ca2 + in cells of 2 groups, 1: 3 group, 37.5μg / 100μl benzoin group and 12.5μg / 100μl group of Suhexiang group were significantly lower than those of the model group The NO content in the group of cells was significantly higher than that in the model group (P <0.05). CONCLUSION: Resuscitation and combination of musk and borneol can effectively protect PC12 cells injured by glucose and hypoxia, which may be related to the increase of NO content in cell supernatant and the decrease of intracellular calcium influx and the decrease of intracellular calcium overload .
