目的研究白细胞介素(IL)1α在人类肝细胞肝癌(HCC)发生中的调节作用。方法收集80例男性和36例女性患者乙型肝炎病毒(HBV)相关的HCC组织、相应的癌旁组织以及正常组织标本。用实时荧光定量PCR方法检测IL-1α、雌激素受体(ER)α、IL-6和MyD88的表达。同时检测二乙基亚硝胺(DEN)诱导的小鼠肝癌模型中IL-1α和IL-6的动态表达水平。结果与正常组织比较,IL-1α在男性癌旁组织中高表达,ERα在男性HCC患者的肿瘤和癌旁组织中都有明显的减少。并且在男性的癌旁组织中,IL-1α的增加和ERα表达的减少之间存在线性相关(r=-0.61,P<0.01)。在DEN诱导的小鼠肝癌模型中,IL-1α在新生肿瘤中高表达,并随着HCC的发生发展而逐渐减少。结论 IL-1α是男性HCC发生发展过程中的一个关键因素,可能是HCC诊断和预测复发的一个有价值的指标。
Objective To investigate the regulatory effect of interleukin (IL) 1α on the development of human hepatocellular carcinoma (HCC). Methods Hepatitis B virus (HBV) -related HCC tissues, corresponding paracancerous tissues and normal tissue specimens from 80 male and 36 female patients were collected. The expression of IL-1α, estrogen receptor (α), IL-6 and MyD88 were detected by real-time fluorescence quantitative PCR. The dynamic expression levels of IL-1α and IL-6 in mouse hepatocellular carcinoma induced by diethylnitrosamine (DEN) were also detected. Results Compared with normal tissues, IL-1α was overexpressed in paracancerous tissues of male patients, and ERα was significantly reduced in both tumor and paracancerous tissues of male patients with HCC. And there was a linear correlation between the increase of IL-1α and the decrease of ERα expression in the paracancerous tissues of males (r = -0.61, P <0.01). In the DEN-induced mouse liver cancer model, IL-1α is highly expressed in neoplastic tumors and gradually decreases with the development of HCC. Conclusion IL-1α is a key factor in the development and progression of HCC in men and may be a valuable indicator for the diagnosis and prognosis of HCC.
