目的:建立首乌藤中大黄素的反相高效液相色谱法含量测定方法,并对不同产地和采收时间的17个样品进行了定量分析。方法:Agilent zorbax SB-C_(18)column(250 mm×4.6 mm,5μm)甲醇-水-磷酸(780∶220∶1)为流动相,检测波长286 nm。结果:大黄素在4.2~124.8μg·ml~(-1)范围内呈良好线性关系(r=1.000 0)。平均回收率103.3%,RSD为2.2%。结论:方法灵敏快速,准确可靠,可用于首乌藤的质量控制。
Objective: To establish a method for the determination of emodin in Radix Polygoni multiflori by reversed-phase high performance liquid chromatography and quantitative analysis of 17 samples from different producing areas and harvest time. METHODS: Agilent zorbax SB-C_(18) column (250 mm×4.6 mm, 5 μm) methanol-water-phosphoric acid (780:220:1) was used as the mobile phase and the detection wavelength was 286 nm. Results: The emodin showed a good linear relationship (r=1.000 0) in the range of 4.2-124.8 μg·ml -1 . The average recovery rate was 103.3%, and the RSD was 2.2%. Conclusion: The method is sensitive, rapid, accurate and reliable and can be used for quality control of Radix Polygoni multiflori.
