【目的】检测了早期流产、正常妊娠和非妊娠大鼠子宫中白细胞介素-18(Interleukin-18,IL-18)、核转录因子(Nuclear factor-kappa B,NF-кB)及诱导型一氧化氮合酶(Inducible nitric oxide synthase,iNOS)的分布与表达,探讨IL-18、iNOS和NF-кB的表达与流产的关系。【方法】采用阴道涂片法将经过自然交配的大鼠分为流产组、正常妊娠组和非妊娠组3组,每组10只。流产组大鼠于妊娠第7天和第8天分别灌服米非司酮4 mg/(kg.d)。各实验组大鼠均于交配后第9天处死,取子宫,切片,免疫组织化学SP法染色后进行图像分析。【结果】IL-18、NF-кB和iNOS在正常妊娠组、非妊娠组和流产组大鼠子宫腔上皮细胞、腺上皮细胞、子宫基质细胞或蜕膜细胞、子宫肌层平滑肌细胞及部分血管内皮细胞中均有表达。与正常妊娠组相比,流产组大鼠子宫中IL-18和iNOS的表达极显著减弱(P<0.01),NF-кB表达极显著增强(P<0.01),血管内皮细胞和平滑肌细胞中IL-18、NF-кB和iNOS的表达均极显著增加(P<0.01)。【结论】IL-18、NF-кB和iNOS的异常表达可能与流产有关。

Abortion group rats were given mifepristone 4 mg / (kg.d) on the 7th day and the 8th day of pregnancy, respectively. The rats in each experimental group were sacrificed on the 9th day after mating and the uterus and sections were taken for immunohistochemical SP staining. 【Results】 IL-18, NF-кB and iNOS expression in uterine cavity epithelial cells, glandular epithelial cells, uterine stromal cells or decidual cells, myometrial smooth muscle cells and some blood vessels in normal pregnancy group, non-pregnant group and abortion group Endothelial cells are expressed. Compared with the normal pregnancy group, the expression of IL-18 and iNOS in the uterus of abortion group was significantly decreased (P <0.01) and the expression of NF-кB was significantly increased (P <0.01), IL -18, NF-кB and iNOS expression were significantly increased (P <0.01). 【Conclusion】 Abnormal expression of IL-18, NF-κB and iNOS may be related to abortion.
