目的观察地塞米松对体外培养人羊膜上皮WISH细胞糖皮质激素受体(GR)-α表达的调节。方法将体外培养的WISH细胞分为不同浓度地塞米松(10-8、10-7、10-6、10-5、10-4mol/L)作用48h组,和单一浓度地塞米松作用不同时间组(终浓度10-6mol/L,培养24、487、2h)。两组均设空白对照组。采用RT-PCR方法检测各组细胞中GR-αmRNA的表达。结果在不同浓度地塞米松作用48h后,10-4mol/L组细胞GRα mRNA表达强于空白组,其余各组细胞GR-α mRNA表达均弱于与空白组(P<0.01)。在地塞米松作用不同时间组,各亚组细胞GR-α mRNA表达与空白组相比,均明显减弱(P<0.01)。结论低浓度地塞米松可下调WISH细胞中GR-αmRNA表达;高浓度地塞米松可上调GRα mRNA的表达。GR-α mRNA表达不随地塞米松作用时间的延长而下降。临床上常规剂量和疗程应用地塞米松促胎肺成熟时,不会导致促分娩发动作用。
Objective To observe the regulation of dexamethasone on the expression of glucocorticoid receptor (GR) -α in cultured human amniotic epithelial WISH cells. Methods WISH cells cultured in vitro were divided into different groups of dexamethasone (10-8, 10-7, 10-6, 10-5 and 10-4 mol / L) for 48h, and treated with dexamethasone at different concentrations for different time Group (final concentration 10-6mol / L, culture 24,487,2h). Two groups were set up blank control group. The expression of GR-αmRNA in each group was detected by RT-PCR. Results After treated with dexamethasone for 48h, the expression of GRα mRNA in 10-4mol / L group was stronger than that in blank group. The expression of GR-α mRNA in the other groups was weaker than that in blank group (P <0.01). Compared with the blank group, the expression of GR-α mRNA in all subgroups was significantly decreased in dexamethasone treated groups at different time points (P <0.01). Conclusion Low concentration of dexamethasone can down-regulate the expression of GR-αmRNA in WISH cells. High concentration of dexamethasone can up-regulate GRα mRNA expression. The expression of GR-α mRNA did not decrease with dexamethasone prolongation. Clinical routine dose and duration of dexamethasone to promote fetal lung maturity, will not lead to promote childbirth start.
