目的研究白介素2α受体(IL-2Rα)在人视网膜色素上皮(RPE)细胞中的表达及白介素2(IL-2)对RPE细胞增生的影响。方法收集培养的2~4代的人胚胎RPE细胞,应用RT-PCR技术,用IL-2α受体特异引物对IL-2α受体进行检测,应用荧光激活细胞分类技术检测IL-2的结合。用抗CD25抗体的免疫荧光染色法识别IL-2α受体的表达,通过3H摄取技术评估重组IL-2对RPE细胞增生的影响。结果RPE细胞可表达IL-2RαmRNA,免疫荧光染色和IL-2结合试验也可表明IL-2α受体的表达,高浓度的IL-2可诱导RPE细胞的增生(P<0·05)。结论培养的RPE细胞能够表达IL-2α受体,重组IL-2可促进RPE细胞的增生。
Objective To investigate the expression of interleukin 2 receptor (IL-2Rα) in human retinal pigment epithelium (RPE) cells and the effect of interleukin-2 (IL-2) on the proliferation of RPE cells. Methods Human embryonic RPE cells from 2 to 4 generations were collected and the IL-2α receptor was detected by RT-PCR and IL-2α receptor specific primers. The binding of IL-2 was detected by fluorescence activated cell sorting. The expression of IL-2α receptor was identified by immunofluorescence staining with anti-CD25 antibody, and the effect of recombinant IL-2 on RPE cell proliferation was evaluated by 3H uptake assay. Results RPE cells could express IL-2RαmRNA. The immunofluorescence staining and IL-2 binding assay also showed that the expression of IL-2α receptor, and the high concentration of IL-2 could induce the proliferation of RPE cells (P <0.05). Conclusion The cultured RPE cells can express IL-2α receptor and recombinant IL-2 can promote the proliferation of RPE cells.
