目的探讨不同浓度17β-雌二醇(17β-E2)对小鼠肺成纤维细胞凋亡及Ⅰ型胶原表达的影响。方法将体外培养的小鼠肺成纤维细胞分为5组:空白对照组,SiO2组,SiO2+不同剂量17β-E2组(10-8、10-7、10-6 mol/L),于作用24、48、72h后收集细胞,应用流式细胞仪检测细胞凋亡,免疫组化法检测Ⅰ型胶原表达。结果与空白对照组比较,SiO2处理组小鼠肺成纤维细胞凋亡率明显降低(P<0.05),而SiO2+17β-E2处理组各时点随着17β-E2剂量增大凋亡率明显升高(P<0.05),随处理时间延长,各剂量17β-E2组成纤维细胞凋亡率明显升高(P<0.05);各处理组Ⅰ型胶原表达有明显差异,与空白对照组比较,SiO2组Ⅰ型胶原表达均明显增高(P<0.05),而17β-E2处理组Ⅰ型胶原表达均明显低于SiO2处理组,差异均有统计学意义(P<0.05)。结论 17β-E2促进小鼠肺成纤维细胞凋亡,抑制Ⅰ型胶原表达,进而抑制肺纤维化。
Objective To investigate the effects of 17β-estradiol (17β-E2) on the apoptosis of lung fibroblasts and the expression of type I collagen in mice. Methods Lung fibroblasts cultured in vitro were divided into 5 groups: blank control group, SiO2 group, SiO2 + different doses of 17β-E2 group (10-8,10-7,10-6 mol / L) After 48 and 72 hours, the cells were harvested, and the apoptosis was detected by flow cytometry. The expression of type Ⅰ collagen was detected by immunohistochemistry. Results Compared with the blank control group, the apoptotic rate of lung fibroblasts in the SiO2-treated group was significantly decreased (P <0.05), while the apoptotic rate of the SiO2-17β-E2 treated group increased with the increase of 17β-E2 (P <0.05). With the prolongation of treatment time, the apoptotic rate of fibroblasts in 17β-E2 group was significantly increased (P <0.05). Compared with the blank control group, The expression of type I collagen in SiO2 group was significantly increased (P <0.05), while the expression of type I collagen in 17β-E2 group was significantly lower than that in SiO2 group (P <0.05). Conclusion 17β-E2 can promote lung fibroblast apoptosis, inhibit the expression of type Ⅰ collagen, and then inhibit pulmonary fibrosis.
