目的:建立超高效液相色谱(UPLC)同时测定醉鱼草Buddleja lindleyana茎、叶、花、果实中蒙花苷、木犀草素、金合欢-7-O-β-D-葡萄糖苷、金合欢素4种黄酮类成分含量方法,比较在醉鱼草各个部位中4种成分的分布情况。方法:采用Waters Acquity BEH C18色谱柱(2.1 mm×50 mm,1.7μm),流动相乙腈-0.1%甲酸水溶液(梯度洗脱),流速0.15 m L·min-1,检测波长330 nm,柱温30℃,进样体积2μL。结果:蒙花苷、木犀草素、金合欢-7-O-β-D-葡萄糖苷和金合欢素的进样量分别在0.002 0~0.600 3μg(r=0.999 7),0.000 4~0.040 1μg(r=0.999 9),0.000 4~0.039 9μg(r=0.999 9),0.000 2~0.020 0μg(r=0.999 9)和峰面积呈良好的线性关系,仪器精密度及方法重复性均良好,符合定量测定要求。平均回收率分别为100.2%,97.1%,94.3%,92.4%,RSD分别为0.7%,1.1%,2.3%,1.9%。结论:4种黄酮类成分含量在醉鱼草不同部位存在较大差异,其中醉鱼草花中4种黄酮总含量最高,通过比较各不同部位成分的含量差异,为醉鱼草的进一步开发利用提供实验依据。该检测方法操作简单,分离效果好,灵敏度高,可用于醉鱼草不同部位中多个黄酮成分的含量测定。
OBJECTIVE: To establish a method for the simultaneous determination of melphalan, luteolin, acacia-7-O-β-D-glucoside in stem, leaves, flowers and fruits of Buddleja lindleyana by ultra performance liquid chromatography 4 flavonoids content of the method of comparison, in each part of Buddleja four components in the distribution. METHODS: Waters Acquity BEH C18 column (2.1 mm × 50 mm, 1.7 μm) was used with a mobile phase of acetonitrile-0.1% formic acid (gradient elution) at a flow rate of 0.15 mL · min- 30 ℃, injection volume 2μL. Results: The injection volume of monolin, luteolin, acacia-7-O-β-D-glucoside and acacetin were respectively 0.002 0-0.600 3 μg (r = 0.999 7), 0.000 4-0.040 1 μg (r = 0.999 9), 0.000 4 ~ 0.039 9 μg (r = 0.999 9), 0.000 2 ~ 0.020 0 μg (r = 0.999 9) and peak area showed a good linear relationship, the instrument precision and method repeatability were good, in line with Quantitative determination of requirements. The average recoveries were 100.2%, 97.1%, 94.3% and 92.4%, respectively, with RSDs of 0.7%, 1.1%, 2.3% and 1.9%, respectively. Conclusion: The contents of four flavonoids are different in different parts of Buddleia, and the content of the four flavonoids in Buddleray flower is the highest. By comparing the content differences of the components in different parts, we provide the further development and utilization of Buddleia Experimental basis. The method has the advantages of simple operation, good separation effect and high sensitivity, and can be used for determining the content of a plurality of flavone components in different parts of Buddleia.
