目的:探讨环境有机污染物多氯联苯暴露对斑马鱼生存率、脊柱形态及骨形态发生蛋白(bone morphogenetic pro-tein,BMP)-2、BMP-4基因表达的影响。方法:以斑马鱼为模式动物,配制0.125、1.000 mg/L 2个不同浓度的多氯联苯对斑马鱼胚胎进行暴露处理,同时设空白对照,观察多氯联苯暴露对斑马鱼幼鱼存活率、脊柱形态及BMP-2、BMP-4基因表达的影响。结果:①低浓度(0.125 mg/L)多氯联苯暴露120 hpf对斑马鱼幼鱼的存活率、脊柱形态无明显影响;②高浓度(1 mg/L)多氯联苯暴露120 hpf后,斑马鱼存活率明显下降且存活的斑马鱼大多出现了脊柱弯曲畸形的现象;③低、高浓度的多氯联苯暴露120 hpf后BMP-2及BMP-4基因表达均较空白对照组明显降低(P<0.05)。结论:不同浓度的多氯联苯暴露对斑马鱼骨骼发育均有一定的毒害作用,提示我们必须加强对多氯联苯等环境有机污染物的监测和控制。
Objective: To investigate the effects of environmental exposure to PCBs on the survival rate, spine morphology and the expression of bone morphogenetic pro-tein (BMP) -2 and BMP-4 in zebrafish. Methods: The zebrafish was used as animal model to prepare zebrafish embryos exposed to different concentrations of PCBs (0.125 and 1.000 mg / L). Blank control was also established to observe the survival of zebrafish Rate, spinal morphology and BMP-2, BMP-4 gene expression. Results: (1) Exposure to low concentrations (0.125 mg / L) of PCBs for 120 hpf had no significant effect on the survival rate and spine morphology of zebrafish juveniles; ② After exposure to 120 hpf of PCBs exposed to high concentrations (1 mg / L) , The zebrafish survival rate decreased significantly and most of the surviving zebrafish showed the phenomenon of spinal deformity. ③ The mRNA expression of BMP-2 and BMP-4 in low and high concentration PCBs was significantly higher than that in the blank control group after 120 hpf exposure Decreased (P <0.05). Conclusion: Exposure to different concentrations of PCBs has some toxic effects on the development of zebrafish skeleton, suggesting that we must strengthen the monitoring and control of PCBs and other environmental organic pollutants.
