用聚合酶链反应(PCR)获得抗-HBs单克隆抗体Fv段轻、重链基因,经连接后形成一条单链含轻、重链的Fv基因(ScFv),并在重组-噬菌体-抗体系统(RPAS)中获得表达,用酶联免疫吸附法(ELISA)证明表达的Fv段具有特异性抗原结合活性。
Polymerase chain reaction (PCR) was used to obtain the light and heavy chain genes of the anti-HBs monoclonal antibody Fv, which were ligated to form a single chain light and heavy chain containing the Fv gene (ScFv) and expressed in a recombinant phage-antibody system (RPAS). The expressed Fv fragment was proved to have specific antigen-binding activity by enzyme-linked immunosorbent assay (ELISA).
