目的:探讨运动预适应是否下调帕金森小鼠中脑细胞bax/bcl-2mRNA表达比值。方法:32只雄性小鼠(8~9周龄)首先随机分为安静组、运动组。运动组连续5周,每天1次中等强度(12米/分,20分钟)跑台训练。然后以上两组再各自随机分成两组,于训练结束后第2天分别注射生理盐水或中等剂量MPTP(30mg/kg×2次)。只有表现典型行为变化的MPTP小鼠被认为是成功帕金森模型小鼠,进入最终分组,分为安静+生理盐水组(Se+Sa)、运动+生理盐水组(E+Sa)、安静+MPTP组(Se+M)、运动+MPTP组(E+M)4组,每组8只。训练结束后第11天处死动物,检测小鼠中脑bax、bcl-2mRNA表达,计算bax/bcl-2mRNA表达比值。结果:E+M组小鼠中脑baxmRNA表达较Se+M组显著降低(P<0.05),E+M组bcl-2mRNA表达较Se+M组显著增加(P<0.05),E+M组bax/bcl-2比值显著小于Se+M组(P<0.05)。结论:运动预适应改变帕金森小鼠中脑黑质细胞凋亡信号分子bcl-2、baxmRNA表达,降低bax/bcl-2mRNA表达比值。
Objective: To investigate whether exercise preconditioning down-regulated the ratio of bax / bcl-2 mRNA in brain cells of Parkinson’s disease mice. Methods: Thirty-two male mice (8-9 weeks old) were randomly divided into quiet group and exercise group. Exercise group for 5 weeks, 1 times a day moderate intensity (12 m / min, 20 minutes) treadmill training. The above two groups were then randomly divided into two groups. Normal saline or moderate dose MPTP (30mg / kg × 2) was injected on the second day after training. Only MPTP mice showing typical behavioral changes were considered to be successful Parkinson’s model mice, which entered the final group and divided into Se + Sa group, exercise + saline group (E + Sa), Quiet + MPTP Group (Se + M), exercise + MPTP group (E + M) 4 groups with 8 rats in each group. Animals were sacrificed on the 11th day after training. The bax and bcl-2 mRNA expressions in the midbrain of the mice were measured, and the ratio of bax / bcl-2 mRNA expression was calculated. Results: The expression of bax mRNA in midbrain of E + M group was significantly lower than that of Se + M group (P <0.05), while the expression of bcl-2 mRNA in E + M group was significantly higher than that of Se + M group (P <0.05) bax / bcl-2 ratio was significantly lower than Se + M group (P <0.05). Conclusion: Exercise preconditioning can change the expression of bcl-2, bax mRNA and the ratio of bax / bcl-2 mRNA in substantia nigra neurons in Parkinson mice.
