目的研究常氧与缺氧条件下慢性牙周炎大鼠血清中基质金属蛋白酶2(matrixmetalloproteinase-2,MMP-2)的表达变化,探讨MMP-2与高原牙周病的关系。方法模拟高原缺氧建立大鼠慢性牙周炎模型,采用ELISA法测定正常对照组、常氧牙周炎组、缺氧对照组、缺氧牙周炎组大鼠血清中MMP-2表达。结果正常对照组、常氧牙周炎组、缺氧对照组、缺氧牙周炎组血清中MMP-2浓度分别为(92.00±21.78)、(118.09±25.01)、(102.03±21.30)、(159.19±34.13)ng/ml,常氧牙周炎组与正常对照组、缺氧牙周炎组与缺氧对照组、缺氧对照组与正常对照组、缺氧牙周炎组与常氧牙周炎组比较均有统计学意义(P<0.01)。其中缺氧牙周炎组血清中MMP-2浓度升高最为显著(P<0.01)。结论缺氧上调血清中MMP-2的表达,进而促进大鼠牙周组织的破坏。
Objective To investigate the expression of matrix metalloproteinase-2 (MMP-2) in the serum of rats with chronic periodontitis under normoxia and hypoxia, and to explore the relationship between MMP-2 and plateau periodontal disease. Methods The model of chronic periodontitis in rats was established by hypoxia on the plateau. The serum levels of MMP-2 in the normal control group, the normoxia periodontitis group, the hypoxia control group and the hypoxia periodontitis group were determined by ELISA. Results The serum levels of MMP-2 in normal control group, normoxia periodontitis group, hypoxia control group and hypoxia periodontitis group were (92.00 ± 21.78), (118.09 ± 25.01), (102.03 ± 21.30), 159.19 ± 34.13) ng / ml, normal periodontitis group and normal control group, hypoxic periodontitis group and hypoxia control group, hypoxic control group and normal control group, hypoxic periodontitis group and normoxia group Zhou Yan group were statistically significant (P <0.01). The serum MMP-2 concentration in hypoxia periodontitis group was the highest (P <0.01). Conclusion Hypoxia can up-regulate the expression of MMP-2 in serum and promote the destruction of periodontal tissues in rats.
