目的研究大鼠实验性肝硬变晚期腹水形成与TNFα及肠源性内毒素血症的关系,探讨肝硬变腹水形成的机制.方法以大鼠为实验对象,分为正常对照组(n=8)、肝硬变对照组(n=12)和肝硬变伴腹水组(n=18).实验组采用复合因子复制肝硬变动物模型.各组均测定腹主动脉血中TNFα和内毒素水平,肝硬变组还测定了腹水量.结果随着肝硬变形成,大鼠血中TNFα水平(μg/L)增高,各组含量分别为908±107和958±128(肝硬变组和肝硬变伴腹水组).相关分析表明肝硬变组腹水量与血中TNFα成正相关(r=086,P<005,内毒素水平增高,并与TNFα的浓度成正相关(r=075,P<005).结论肝硬变腹水形成与血中TNFα升高相关,而TNFα的升高则源于肠源性内毒素血症的形成.
Objective To study the relationship between the formation of ascites and TNFα and enterogenic endotoxemia in rats with experimental cirrhosis and to explore the mechanism of cirrhosis with ascites. Methods The rats were divided into normal control group (n = 8), cirrhosis control group (n = 12) and cirrhosis with ascites group (n = 18). The experimental group used compound factor to replicate cirrhosis animal model. The levels of TNFα and endotoxin in abdominal aorta blood were determined in each group, and the amount of ascites was also measured in cirrhotic group. Results With the formation of liver cirrhosis, the level of TNFα (μg / L) in rat blood increased, the contents of each group were 908 ± 107 and 958 ± 128 (cirrhosis group and cirrhosis With ascites group). Correlation analysis showed that the amount of ascites in cirrhotic patients was positively correlated with TNFα in blood (r = 086, P <005, the level of endotoxin was increased and positively correlated with the concentration of TNFα (r = 075, P <0 ⑤ 05) .Conclusion The formation of cirrhosis with ascites is related to the increase of TNFα in blood, whereas the increase of TNFα is due to the formation of enterogenous endotoxemia.
