榄香烯是从中药莪术中提取的抗肿瘤活性物质,对恶性积液具有较好临床疗效,但其抗肿瘤作用机理尚不十分清楚.本文就榄香烯对恶性渗出液相关淋巴细胞(EAL)的作用以及对IL-2诱导EAL增殖和产生抗肿瘤活性的影响进行了实验研究.EAL取自8例病人的恶性胸水,原发灶为肺癌6例,卵巢癌2例,治疗方法为:榄香烯400mg胸腔灌注每周2次,连用3周,疗效为62%(5/8).实验方法为:取治疗前、后的胸水200ml离心沉淀,取细胞沉淀做细胞涂片、HE染色进行细胞学观察,确定治疗前、后胸水中EAL及癌细胞数量变化;以Ficoll-Hypague密度梯度离心法分离EAL,然后用含500U/ml IL-2的培养液将EAL培养72小时,收集EAL用于LAK活性检测.LAK活性检测采用MTT法,靶细胞为Hela细胞,效应细胞取自榄香烯治疗前、后胸水中的EAL,这2种
Elemene is an anti-tumor active substance extracted from traditional Chinese medicine, and has good clinical efficacy for malignant effusion. However, its anti-tumor mechanism is not yet clear. This paper deals with leukemia in malignant exudate-associated lymphocytes ( The effects of EAL) and its effects on IL-2 induced EAL proliferation and antitumor activity were studied. EAL was obtained from 8 patients with malignant pleural effusion. The primary tumor was 6 cases of lung cancer and 2 cases of ovarian cancer. The treatment was : Elemene 400mg thoracic cavity perfusion 2 times a week for 3 weeks, the curative effect was 62% (5/8). The experimental method is: take the pleural effusion 200ml before and after treatment, centrifuge and sediment, take the cell pellet and do the cell smear, HE Staining was performed for cytological observation to determine the number of EAL and cancer cells in the pleural fluid before and after treatment; EAL was isolated by Ficoll-Hypague density gradient centrifugation, and then EAL was cultured in a medium containing 500 U/ml IL-2 for 72 hours. EAL was used for LAK activity detection. LAK activity was measured by MTT assay. The target cells were HeLa cells. The effector cells were taken from the EAL in the pleural fluid before and after treatment with elemene.
