[目的]通过差异蛋白组学方法分析疣状胃炎与同遗传背景胃癌组织的差异蛋白表达,旨在筛选该型胃炎向胃癌转变过程中关键的蛋白分子,探讨其癌变的分子机制。[方法]外科手术获取内镜及病理检查提示同时存在胃癌与疣状胃炎病变患者的病变组织,采用比较蛋白组学通用的双向电泳及质谱分析技术,筛选并鉴定胃癌组织及癌周疣状胃炎组织中差异表达的蛋白质。[结果]胃癌组织与癌旁疣状胃炎组织比较,共筛选出显著差异表达蛋白点34个,其中表达上调蛋白质点20个,表达下调蛋白点14个;共有27个蛋白点被鉴定,上调蛋白17个,下调蛋白10个。[结论]在疣状胃炎基础上发生的胃癌与其疣状胃炎病损组织的蛋白质表达存在显著差异,这些差异表达蛋白分子涉及细胞增殖、凋亡、运动、物质能量代谢等诸多功能领域,其代表的分子信号转导途径可能揭示疣状胃炎癌变的分子机制。
[Objective] To analyze the differential protein expression of verrucous gastritis and gastric cancer tissue with the same genetic background by differential proteomics, and to screen the key protein molecules in the process of gastric gastritis and to discuss the molecular mechanism of carcinogenesis. [Methods] Obtaining endoscopic and pathological examination surgically revealed the presence of lesions in both gastric cancer and verrucous gastritis patients. Two-dimensional gel electrophoresis and mass spectrometry were used to screen and identify gastric cancer and verrucous gastritis Differentially expressed proteins in tissues. [Results] There were 34 differentially expressed protein spots in gastric cancer tissues compared with the adjacent verrucous gastritis tissues, of which 20 were upregulated and 14 were downregulated. A total of 27 protein spots were identified and the proteins upregulated 17, down 10 protein. [Conclusion] There are significant differences in the protein expression of gastric cancer and verrucous gastritis lesions on the basis of verrucous gastritis. The differentially expressed protein molecules are involved in many functional fields such as cell proliferation, apoptosis, exercise, energy metabolism and other substances. Molecular signal transduction pathway may reveal the molecular mechanism of verrucous gastritis.
