目的探讨不同质量浓度臭氧水对牙周炎大鼠牙周组织中白细胞介素-6(IL-6)的表达影响。方法本研究于2013年4—8月在佳木斯大学动物实验中心进行。选用70只Wistar雄性大鼠,随机抽取5只作为正常对照组(颈椎脱臼法处死),其余大鼠均建立牙周炎模型。牙周炎模型建立后,随机抽取5只作为牙周炎未治疗组(颈椎脱臼法处死),其余牙周炎大鼠随机分为臭氧水治疗组和对照治疗组进行牙周袋冲洗。运用一般组织学方法观察牙周组织形态变化,并采用免疫组化法检测牙周组织中IL-6含量的变化。结果牙周炎未治疗组的牙周组织中IL-6表达明显高于正常对照组(P<0.05);各治疗组的牙周组织中IL-6表达均不同程度低于牙周炎未治疗组(P<0.05);在各牙周袋冲洗时间段,15 mg/L臭氧水治疗组的牙周组织中IL-6表达均低于5、10 mg/L臭氧水治疗组(P<0.05),但与对照治疗组的差异无统计学意义(P>0.05)。结论臭氧水能降低炎性牙周组织中IL-6的表达,其中质量浓度为15mg/L的臭氧水对其影响更加明显,对牙周炎有一定治疗效果。
Objective To investigate the effects of different concentrations of ozone water on the expression of interleukin-6 (IL-6) in periodontal tissues of rats with periodontitis. Methods The study was conducted at Animal Experimental Center of Jiamusi University from April to August in 2013. A total of 70 Wistar male rats were selected and randomly selected as normal control group (cervical dislocation method), the rest of the rats were established periodontitis model. After establishment of periodontitis model, 5 rats were randomly selected as the periodontitis untreated group (cervical dislocation method was killed), and the other periodontitis rats were randomly divided into ozone water treatment group and control treatment group for periodontal pocket irrigation. Periodontal tissue morphological changes were observed by using histological method. The changes of IL-6 in periodontal tissues were detected by immunohistochemistry. Results The expression of IL-6 in periodontal tissues of periodontitis group was significantly higher than that of normal control group (P <0.05). The expression of IL-6 in periodontal tissues in each periodontal group was lower than that of periodontitis group (P <0.05). The expression of IL-6 in the periodontal pocket was significantly lower than that in the 5 and 10 mg / L ozone water treatment groups (P <0.05) ), But no significant difference with the control group (P> 0.05). Conclusions Ozonated water can reduce the expression of IL-6 in inflammatory periodontal tissue. Ozone water with mass concentration of 15mg / L has a more obvious effect on periodontitis.
