目的检测程序性死亡受体配体-1(PD-L1)及记忆性T细胞在肝囊型包虫病患者CD4+T淋巴细胞中的表达,探讨其在细粒棘球蚴感染免疫逃避中的作用。方法具有包虫活性的肝包虫病患者42例,健康对照20例,取血,分离PBMC,流式细胞术检测PD-L1(CD274+)、CD45RO+及CD4+CD45RO+CD274+细胞比例,并分析PD-L1与CD45RO的相关性。结果肝囊型包虫病患者PBMC中PD-L1、CD45RO+及CD4+CD45RO+CD274+细胞比例均显著高于健康对照组(P<0.05);肝囊型包虫病患者的PD-L1与CD45RO呈显著正相关(P<0.05),健康对照组二者无显著相关关系(P>0.05)。结论在肝囊型包虫病患者的免疫反应中,PD-L1可能通过记忆性T淋巴细胞发挥促进包虫免疫逃避的作用。
Objective To detect the expression of PD-L1 and memory T cells in CD4 + T lymphocytes of patients with hepatic cystic echinococcosis (ESCC), and to investigate the role of PD-L1 in immune escape of Echinococcus granulosus Role. Methods Forty-two patients with hydatidosis and 20 healthy controls were enrolled in this study. PBMCs were isolated from peripheral blood and the percentage of PD-L1 (CD274 +), CD45RO + and CD4 + CD45RO + CD274 + cells was measured by flow cytometry -L1 and CD45RO. Results The percentage of PD-L1, CD45RO + and CD4 + CD45RO + CD274 + cells in PBMC from patients with hepatic cystic echinococcosis were significantly higher than those from healthy controls (P <0.05). PD-L1 and CD45RO in patients with hepatic cystic echinococcosis (P <0.05). There was no significant correlation between the two groups (P> 0.05). Conclusion In the immune response of patients with hepatic cystic echinococcosis, PD-L1 may play an important role in promoting immune evasion by memory T lymphocytes.
