目的:探讨接骨膏在促进骨折愈合过程中与血生化指标及降钙素基因相关肽(CGRP)表达的关系。方法:将胫骨骨折模型建立成功的大鼠随机分为5组:空白对照组、模型对照组、阳性对照组、接骨膏低剂量(L,1 g·ml~(-1),以生药材计)组和高剂量(H,2 g·ml~(-1),以生药材计)组,每组60只。空白对照组和模型对照组给予2 ml生理盐水,阳性对照组给予2 ml续断骨伤合剂浓缩液,接骨膏L组和H组分别给予2 ml低浓度和高浓度浓缩液,均灌胃给药,qd,连续给药28 d。骨折后第7,14,21,28天取血样本和骨痂标本,进行血生化指标检测、HE染色和免疫组织化学染色,骨折标本行数字化X射线(DR)摄片。结果:与模型对照组比较,接骨膏H组的血清钙磷离子浓度乘积在14,21 d显著升高,血清AKP在14~28 d显著升高,差异有统计学意义(P<0.05)。骨折21 d内接骨膏H组CGRP表达程度与模型对照组比较显著增强,差异有统计学意义(P<0.05)。接骨膏H组与阳性对照组比较,差异无统计学意义(P>0.05)。结论:接骨膏可以通过提高骨折愈合中血清AKP的活性、升高钙磷乘积以及调控神经肽CGRP活性的机制,达到促进骨折愈合的目的。
OBJECTIVE: To investigate the relationship between Jiegu Paste and blood biochemical parameters and the expression of calcitonin gene-related peptide (CGRP) in the process of promoting fracture healing. Methods: The rats with successful tibial fracture model were randomly divided into 5 groups: control group, model control group, positive control group, low dose of Ligu Gao (L, 1 g · ml -1) ) And high dose (H, 2 g · ml -1, raw herbs) group, 60 rats in each group. 2 ml of normal saline was given to the blank control group and the model control group, and 2 ml of the Dioscoreae Granule Mixing Concentrate was given to the positive control group. Ligustrazine group and H group were given 2 ml of low concentration and high concentration respectively, Medicine, qd, continuous administration 28 d. Blood samples and callus samples were taken on the 7th, 14th, 21st and 28th day after fracture. Blood biochemical detection, HE staining and immunohistochemical staining were performed. The digitalized X-ray (DR) radiographs were taken on the fracture specimens. Results: Compared with the model control group, the product of serum calcium and phosphorus ion concentration in the group of Jieu Gao H increased significantly at 14 and 21 d, and the serum AKP increased significantly at 14 and 28 d (P <0.05). The level of CGRP expression in H group was significantly higher than that in the model control group on 21 d after fracture (P <0.05). Compared with the positive control group, there was no significant difference (P> 0.05). Conclusion: Jiegu Paste can promote the fracture healing by improving the activity of serum AKP in fracture healing, increasing the product of calcium and phosphorus and regulating the activity of neuropeptide CGRP.
