本文采用大鼠静脉自身给药实验法评价甲基苯丙胺及其衍生物安啡拉酮的精神依赖性潜力。在自身给药形成实验中,当甲基苯丙胺剂量分别0.25、0.125、0.0625mg·kg-1(注射)时,大鼠在固定比率程序控制下,可以对甲基苯丙胺形成稳定的自身给药反应,有剂量效应关系;当安啡拉酮日剂量相当于人剂量的2.0、5.0、7.0倍时,大鼠对安啡拉团仅产生比盐水稍高的反应率,不稳定,且无剂量效应关系。在替代实验中,大鼠先形成稳定的甲基苯丙胺自身给药行为,安啡拉酮(日剂量相当于人剂量的2-5倍)可以替代甲基苯丙胺维持自身给药反应,有剂量效应关系,但反应率不稳定。在累进比率程序控制下,甲基苯丙胶的自身给药反应“终止点”随剂量的增加而增大。实验表明甲基苯丙胺有较强的精神依赖性潜力,而安啡拉酮较弱。
