本文利用Sephadex LH20凝胶过滤法分离结合和游离的甾体,以定量人子宫内膜胞浆中的孕酮受体(PRc),并同时采用葡聚糖-活性炭吸附法(DCC)对标本进行配对测定。这两种方法测得的特异性结合量(Bs)以及总结合比率(Bt/T)之间,经统计学处理显示有密切正相关存在。LH20法是一种有效的和较为温和的分离测定受体的方法。由于LH20法能直接测定游离甾体浓度,故有利于回收率的精确测定。结果表明,采用LH20法测得的PRc的结合参数可能更为可靠。
In this paper, the bound and free steroids were separated by Sephadex LH20 gel filtration to quantify the progesterone receptor (PRc) in the human endometrial cytoplasm. At the same time, the samples were subjected to dextran-activated carbon adsorption (DCC) Matching test. The specific binding amount (Bs) and the total binding ratio (Bt / T) measured by these two methods show a close correlation with each other statistically. The LH20 method is a potent and milder method for the separation of receptors. Because LH20 method can directly measure free steroid concentration, it is conducive to the accurate determination of recovery rate. The results show that the binding parameters of PRc measured by the LH20 method may be more reliable.
