目的观察Smad4和转化生长因子(TGF)-β1在膀胱癌组织内的表达情况,分析Smad4和TGF-β1的表达与膀胱癌进展及预后之间的关系。方法选取临床资料完整的膀胱癌病例48例,其中,淋巴结转移组20例,无淋巴结转移组28例。免疫组化法检测膀胱癌组织内Smad4和TGF-β1的表达。取膀胱癌术后组织16例,Western blot法检测Smad4和TGF-β1在膀胱癌组织内的表达情况。结果 Smad4表达于肿瘤细胞浆和胞核,在有淋巴结转移膀胱癌组织内的表达量明显低于其在无淋巴结转移组的表达量,Smad4表达与膀胱癌临床分期、分级、淋巴结转移以及膀胱癌复发密切相关(<0.05)。TGF-β1表达于肿瘤细胞的胞浆内,在有淋巴结转移膀胱癌组织中的表达量高于其在无淋巴结转移膀胱癌组织内的表达量,TGF-β1的表达与膀胱癌临床分期、分级及淋巴结转移之间具有相关性(<0.05)。Smad4和TGF-β1在膀胱癌组织内的表达呈显著负相关(r=-0.324,=0.025)。Kaplan-Meier分析表明Smad4阳性患者的五年总体生存率高于Smad4表达阴性患者的生存率,而TGF-β1的表达与膀胱癌患者的预后无关。结论 Smad4和TGF-β1在膀胱癌组织的表达呈负相关,与膀胱癌临床分期、分级及淋巴结转移相关。
Objective To observe the expression of Smad4 and transforming growth factor-β1 (TGF-β1) in bladder cancer and to analyze the relationship between the expression of Smad4 and TGF-β1 and the progression and prognosis of bladder cancer. Methods Forty-eight cases of bladder cancer with complete clinical data were selected, including 20 cases with lymph node metastasis and 28 cases without lymph node metastasis. Immunohistochemistry was used to detect the expression of Smad4 and TGF-β1 in bladder cancer tissues. Sixteen cases of bladder cancer tissues were collected. The expression of Smad4 and TGF-β1 in bladder cancer tissues was detected by Western blot. Results The expression of Smad4 in cytoplasm and nucleus of the tumor was significantly lower than that in the non-lymph node metastasis group, the expression of Smad4 in bladder cancer with clinical stage, grade, lymph node metastasis and bladder cancer Relapse was closely related (<0.05). The expression of TGF-β1 in the cytoplasm of tumor cells was higher in bladder cancer with lymph node metastasis than that in bladder cancer without lymph node metastasis. The expression of TGF-β1 was correlated with the clinical stage and grade of bladder cancer And lymph node metastasis (<0.05). The expression of Smad4 and TGF-β1 in bladder cancer tissues was negatively correlated (r = -0.324, = 0.025). Kaplan-Meier analysis showed that the overall 5-year survival rate of Smad4-positive patients was higher than that of Smad4-negative patients, while the expression of TGF-β1 was not related to the prognosis of bladder cancer patients. Conclusion The expression of Smad4 and TGF-β1 in bladder cancer tissues is negatively correlated with the clinical stage, grade and lymph node metastasis.
