目的 :探讨丁胺卡那霉素作用于耳蜗系统时毛细胞凋亡现象及其在听力损伤中的作用。方法 :15只豚鼠随机分为 3组 ,各组肌内注射丁胺卡那霉素 40 0mg·kg-1·d-1,分别于用药 1d ,3d ,6d后处死。处死前检测其听脑干反应 (auditorybrainstemresponses,ABR)的变化 ,并利用光镜和TUNEL标记技术检测耳蜗毛细胞发生凋亡的情况。结果 :①豚鼠肌内注射丁胺卡那霉素 1d后耳蜗毛细胞即出现凋亡现象 ,连续用药 3~ 6d ,毛细胞凋亡呈强阳性表达 ;②耳蜗毛细胞凋亡出现的早期豚鼠听力阈值无明显改变 ,连续用药 6d后听力阈值则明显提高。结论 :①丁胺卡那霉素作用于耳蜗毛细胞可引起毛细胞凋亡 ,细胞凋亡是豚鼠耳蜗毛细胞损伤的一条途径 ;②丁胺卡那霉素耳毒性作用机制可能与其引起耳蜗毛细胞凋亡有关
Objective: To investigate the apoptosis of hair cells and its role in hearing impairment induced by amikacin in the cochlear system. Methods: Fifteen guinea pigs were randomly divided into three groups. Each group was injected intramuscularly with amikacin 40 0 mg · kg-1 · d-1 and were sacrificed on day 1, day 3 and day 6, respectively. Auditory brainstem responses (ABR) were measured before sacrifice, and apoptosis of cochlear hair cells was detected by light microscopy and TUNEL labeling. Results ①The apoptosis of cochlear hair cells was observed in guinea pigs after intramuscular injection of amikacin for 1 day. The apoptosis of hair cells was strongly positive 3 to 6 days after continuous administration. ② The early guinea pig hearing cells No significant change in threshold, continuous medication 6d after hearing threshold was significantly improved. Conclusion: (1) The effect of amikacin on cochlear hair cells can induce hair cell apoptosis, which is one of the pathways of hair cell damage in guinea pig cochlea. ② The mechanism of amikacin of amikacin may be related to the mechanism of amikacin induced cochlear hair Apoptosis related
