[目的]探讨氯化汞(Hg Cl2)对SD大鼠体外培养睾丸间质细胞(leydig cell)类固醇激素合成的影响。[方法]不同浓度Hg Cl2(1×10-8、1×10-7、1×10-6、1×10-5 mol/L)作用于leydig细胞后,测定孕酮含量,运用实时荧光定量逆转录聚合酶链反应(RT-PCR)技术检测Hg Cl2对leydig细胞中St AR、P450scc和3β-HSD m RNA表达的影响。[结果]经不同浓度Hg Cl2染毒后,leydig细胞分泌孕酮的量均有所降低,其中1×10-7、1×10-6、1×10-5 mol/L Hg Cl2染毒组与对照组相比,差异均有统计学意义(P<0.05)。1×10-7、1×10-6和1×10-5 mol/L Hg Cl2剂量组的St AR、P450scc和3β-HSD m RNA相对表达量均低于对照组(P<0.05)。[结论]汞可以影响睾丸间质细胞类固醇激素的合成。
[Objective] To investigate the effect of mercuric chloride (Hg Cl2) on the synthesis of leydig cell steroid hormone in SD rats in vitro. [Method] After treated with different concentrations of HgCl2 (1 × 10-8, 1 × 10-7, 1 × 10-6, 1 × 10-5 mol / L) in leydig cells, the content of progesterone was measured. The effect of Hg Cl2 on the expression of St AR, P450scc and 3β-HSD m RNA in leydig cells was detected by reverse transcription-polymerase chain reaction (RT-PCR). [Result] The amount of progesterone secreted by leydig cells decreased after exposure to different concentrations of Hg Cl2, among which 1 × 10-7, 1 × 10-6 and 1 × 10-5 mol / L HgCl2 groups Compared with the control group, the differences were statistically significant (P <0.05). The relative expression levels of St AR, P450scc and 3β-HSD m RNA in 1 × 10-7, 1 × 10-6 and 1 × 10-5 mol / L HgCl2 groups were lower than those in the control group (P <0.05). [Conclusion] Mercury can affect the synthesis of testicular stromal cells steroid hormones.
