目的:观察银杏叶提取物(EGb)对白介素-1β损伤的关节软骨细胞分泌总蛋白的影响。方法:在无菌环境下消化得到3月龄雌性新西兰白兔膝关节软骨细胞,实验用细胞为第2代软骨细胞。软骨细胞与IL-1β(5μg/L)、IL-1β(5μg/L)+地塞米松(10-6、10-5mmol/L)、IL-1β(5μg/L)+EGb(80、160mg/L)共同孵育,分别在72h后考马斯亮蓝检测软骨细胞分泌总蛋白的情况。结果:IL-1β使软骨细胞的总蛋白分泌减少(P<0.01)。EGb可对抗IL-1β的效应,此效应有剂量依赖性(P<0.01)。结论:银杏叶提取物EGb761通过调节受损伤后软骨细胞分泌总蛋白以保护IL-1β损伤的膝关节软骨细胞。
Objective: To observe the effect of Ginkgo biloba extract (EGb) on total protein secreted by articular chondrocytes induced by interleukin-1β. METHODS: Knee chondrocytes of 3-month-old female New Zealand white rabbits were digested in aseptic environment. The experimental cells were passage 2 chondrocytes. Chondrocytes were significantly different from those of IL-1β (5μg / L), IL-1β (5μg / L) + dexamethasone (10-6,10-5mmol / L) / L) co-incubation, respectively, after 72h Coomassie brilliant blue to detect total protein secretion of chondrocytes. Results: IL-1β reduced the total protein secretion of chondrocytes (P <0.01). EGb antagonizes the effect of IL-1β in a dose-dependent manner (P <0.01). Conclusion: Ginkgo biloba extract EGb761 protects articular chondrocytes damaged by IL-1β by regulating the total protein secreted by chondrocytes.
