目的寻求适合O139群霍乱疫情的病原学检测方法,以便为O139群霍乱疫情的防控提供及时可靠的科学依据。方法用实时荧光定量PCR仪进行O139群霍乱弧菌的核酸检测、常规分离培养、O139群霍乱弧菌快速检测(胶体金法)。结果 O139群霍乱弧菌的核酸检测阳性结果9份,且从这9份标本中分离到阳性菌株9株,有7份试样第一次增菌液胶体金法呈阳性反应,2份试样经过第二次增菌后胶体金法才呈阳性反应。结论实时荧光定量PCR法和胶体金法作为辅助检测O139群霍乱弧菌的方法,可以较快得到初步结果,将两种方法与传统的分离培养鉴定相结合,可以保证结果的及时性和准确性。
Objective To search for an etiological detection method suitable for the cholera epidemic of O139 group in order to provide scientific basis for timely and reliable prevention and control of the cholera epidemic of O139 group. Methods The real-time fluorescence quantitative PCR instrument was used to detect the nucleic acid of Vibrio cholerae O139, routinely isolated and cultured, and Vibrio cholerae O139 was detected rapidly (colloidal gold method). Results 9 samples were positive for nucleic acid detection of Vibrio cholerae O139 strains, and 9 strains were positive isolated from these 9 samples. Among them, 7 samples were positive for the first time by bacteria colloidal gold method, 2 samples After the second increase in bacteria colloidal gold law was a positive reaction. Conclusion Real-time fluorescence quantitative PCR and colloidal gold as a method to detect Vibrio cholera O139 group can get preliminary results quickly and combine the two methods with traditional isolation and culture identification to ensure the timeliness and accuracy of the results .
