为研究LNG在大鼠原代培养肝细胞中的代谢过程,采用大鼠肝脏在体灌流技术,分离肝细胞与LNG进行原代培养,分为2.5,7.5μg/ml两组,在不同时间取0.5ml样本,用HPLC法测定LNG浓度。测定第1组K值为0.335±0.10 h~(-1),T_(1/2)为2.069±0.34h;第2组K值为0.16±0.05h~(-1),T_(1/2)为4.33±0.69h。结果显示大鼠肝细胞对LNG有一定的代谢能力,在一定的药物浓度范围内其代谢动力学符合一级动力学过程。
