目的:探讨没药中环阿尔廷烷型三萜化合物环阿尔廷-24-烯-1α,2α,3β-三醇对前列腺肿瘤细胞生长的抑制作用。方法:Hoechst染色法观察细胞形态学变化;流式细胞术检测细胞周期和凋亡的变化;蛋白质印迹分析检测细胞凋亡相关蛋白的表达。结果:经环阿尔廷-24-烯-1α,2α,3β-三醇处理后,PC3细胞核发生凋亡特征性变化,并经流式细胞术证明其能促进PC3细胞凋亡,并能将细胞阻滞于G0/G1期,其中伴随细胞凋亡相关蛋白BCL-2、BAX、Caspase-3和Caspase-9表达水平的变化。结论:环阿尔廷-24-烯-1α,2α,3β-三醇能通过诱导肿瘤细胞凋亡和细胞周期阻滞,抑制人前列腺肿瘤PC3细胞的增殖。
OBJECTIVE: To investigate the inhibitory effect of myrrh Central Aartin alkane type triterpene ring aartin-24-ene-1α, 2α, 3β-triol on the growth of prostate tumor cells. Methods: Hoechst staining was used to observe the changes of cell morphology. Cell cycle and apoptosis were detected by flow cytometry. Western blotting was used to detect the expression of apoptosis related proteins. Results: Apoptotic changes of PC3 cells were observed after treatment with cycloarten-24-ene-1α, 2α, 3β-triol. Flow cytometry demonstrated that PC3 cells could promote the apoptosis of PC3 cells, It was arrested at G0 / G1 phase, which was accompanied by the changes of the expression of apoptosis related proteins BCL-2, BAX, Caspase-3 and Caspase-9. CONCLUSION: Cycloarterine-24-ene-1α, 2α, 3β-triol can inhibit the proliferation of human prostate cancer PC3 cells by inducing tumor cell apoptosis and cell cycle arrest.
