为了检测味觉受体偶联G蛋白Gγ13和α-gustducin基因在雌性生殖系统的表达,探讨2种G蛋白在雌性生殖机能中可能发挥的作用及其在生殖医学方面的潜在应用。用RT-PCR和超敏ABC免疫组化方法,检测Gγ13和α-gustducin在6周龄间情期小鼠卵巢、子宫、输卵管组织中的基因表达和蛋白分布。结果表明,在小鼠雌性生殖系统中均发现Gγ13和α-gustducin基因的转录子,免疫组化研究进一步证实了Gγ13和α-gustducin确实在小鼠卵巢颗粒黄体细胞、卵母细胞、子宫外膜、子宫内膜腺体、子宫肌层、输卵管浆膜和输卵管黏膜处存在阳性分布。本试验表明味觉受体偶联G蛋白Gγ13和α-gustducin在雌性小鼠生殖系统中广泛分布,预示着雌性生殖系统也可能对味觉物质进行感知,为进一步研究味觉受体在雌性生理机能中可能发挥的调控功能奠定初步的研究基础,同时也为味觉的细胞生物学研究提供新方向。
In order to detect the expression of G protein Gγ13 and α-gustducin genes in the female reproductive system, we investigated the possible roles of these two G proteins in female reproductive function and their potential applications in reproductive medicine. The gene expression and protein distribution of Gγ13 and α-gustducin in ovary, uterus and oviduct tissues of 6-week-old estrous mice were detected by RT-PCR and hypersensitive ABC immunohistochemistry. The results showed that both Gγ13 and α-gustducin gene transcripts were found in mouse female reproductive system. Immunohistochemical study further confirmed that Gγ13 and α-gustducin did inhibit ovarian granulosa cells, oocytes and extrauterine membranes , Endometrial glands, myometrium, tubal serous and tubal mucosa at the positive distribution. This experiment shows that the GABA-Gγ13 and α-gustducin are widely distributed in the reproductive system of female mice, suggesting that the female reproductive system may also be able to sense the taste substances.
