目的建立测定松果菊苷、毛蕊花糖苷、异麦角甾苷、2’-乙酰基麦角甾苷的胶束毛细管电泳方法。方法高效毛细管电泳仪采用PDA检测器,紫外检测波长为214 nm,分离使用内壁未涂覆的石英毛细管,60.2 cm×75.0μm,有效长度为50.0 cm。研究运行电解质及胶束浓度对分析的影响。结果在100mmol/L硼砂、40mmol/L十二烷基硫酸钠,18kV的胶束毛细管电泳分离电压条件下,实现了对上述活性分析物的有效分离。结论此胶束毛细管电泳方法准确、简单、快速,可用于新疆产管花肉苁蓉中上述活性成分的测定及肉苁蓉的质量控制。
Objective To establish a micellar capillary electrophoresis method for the determination of echinacoside, verbascoside, isoacteoside and 2’-acetyl acteoside. Methods High performance capillary electrophoresis with PDA detector was used. The UV detection wavelength was 214 nm. The uncoated silica capillary was used for the separation. The effective length was 60.2 cm × 75.0 μm and the effective length was 50.0 cm. The effect of running electrolyte and micellar concentration on the analysis was studied. Results The effective separation of the above analytes was achieved under the conditions of 100 mmol / L borax, 40 mmol / L sodium dodecyl sulfate and 18 kV micellar capillary electrophoresis separation voltage. Conclusions The micellar capillary electrophoresis method is accurate, simple and rapid and can be used for the determination of the above active ingredients in Cistanche tubulosa and the quality control of Cistanche tubulosa in Xinjiang.
