目的 测定异丙酚全血浓度。方法 抗凝全血经环己烷提取后,氮气吹干。残余物用流动相溶解后由高效液相色谱仪进行测定。流动相为乙腈- 双蒸水- 三氟醋酸(40∶60∶0.1) 。荧光检测器,激发波长276 nm ,发射波长310 nm 。结果 该方法的最低检测浓度为25 μg·L-1 。日内相对标准偏差=4 .1% ,日间相对标准偏差=4.0% . 回收率为90 .3 % 。结论 本方法检测异丙酚全血浓度,灵敏度、精密性和稳定性均较高,实用性强。
Objective To determine the concentration of propofol whole blood. Methods Anticoagulated whole blood was extracted with cyclohexane and then blown dry with nitrogen. The residue was dissolved in the mobile phase and then measured by a high-performance liquid chromatograph. The mobile phase was acetonitrile-double distilled water-trifluoroacetic acid (40:60:0.1). Fluorescence detector, excitation wavelength 276 nm, emission wavelength 310 nm. Results The lowest concentration of this method was 25 μg · L-1. Days relative standard deviation = 4. 1%, daytime relative standard deviation = 4.0%. The recovery rate is 90. 3%. Conclusion The method for the determination of propofol whole blood concentration, sensitivity, precision and stability are high, practicality.
