目的分析胃腺癌BGC823细胞Zbtb7A高表达前后差异基因的表达。方法应用22K人基因组寡核苷酸芯片,筛选BGC823细胞高表达Zbtb7A前后差异表达的基因,采用RT-PCR验证部分差异基因的表达。结果发现Zbtb7A高表达前后BGC823细胞有显著性表达差异的基因共312个,其中上调基因205个,下调基因107个。经RT-PCR验证部分差异基因的表达与芯片结果一致。结论 Zbtb7A高表达前后胃腺癌BGC823细胞部分功能基因存在着显著的表达差异,多数差异表达的基因参与细胞黏附、细胞骨架形成以及细胞代谢等过程。
Objective To analyze the differential gene expression of Zbtb7A in gastric adenocarcinoma BGC823 cells before and after high expression. Methods 22K human genome oligonucleotide microarray was used to screen the differentially expressed genes of BGC823 cells before and after high expression of Zbtb7A, and RT-PCR was used to verify the expression of some of the differentially expressed genes. The results showed that there were 312 genes with significant difference between BGC823 cells and Zbtb7A before and after high expression, of which 205 were up-regulated and 107 were down-regulated. The result of RT-PCR showed that the expression of some differentially expressed genes was consistent with the chip results. Conclusion There were significant differences in some functional genes between BGC823 cells and Zbtb7A before and after high expression of Zbtb7A. Most differentially expressed genes involved in cell adhesion, cytoskeleton formation and cell metabolism.
